ADDENDUM REPORT
Follow-up cytoegentic assessment of the parental karyotypes reveals that the deletion is a de novo (dn) rearrangement.
CYTOGENETIC REPORT
Indications for Testing
Intregrated screening indicates elevated risk (1/65 for Down and 1/30 for Edward's Syndrome)
Methods and Analysis
Amniocytes were cultured in situ by standard methods
Number colonies assessed: 13
Metaphases were stained by standard trypsin-Giemsa G-banding methods
Level of resolution achieved: approximately 400 bands per haploid set
Metaphase fluorescence in situ hybridization (ish) was performed using commercially available "research" probes. The signal patterns were assessed in 6 metaphass.
Karyotype and Interpretation
46,XX,del(8) (p23.1), ish del (8) (p23.1)(8ptel-,8qtel+)
The modal karyotypeis an abnormal, unbalanced female karyotype. All metaphases assessed have a chromosomal anomaly that we interpret, with present information, as a terminal deletion of the short arm of chromosome 8, with breakpoint estimated at the junction of bands 8p23.1 and 8p23.2 (segment deleted is 8p23.2-8pter). FISH studies with probes specific for the subtelomeric regions of chromosome 8 were performed. The results support our interpretation by conventional cytogenetic.
Genetic counselling is strongly recomended so that the significance of this finding, its implications and follow-up options might be discussed. Please note that a complete investigation of this finding includes but may ot be limited to a parental cytogenetic assessment in the light of the obsrvations reported.
